Male-pattern baldness may be the effect of a protein in the scalp, according to recent research that raises the possibility that drugs being tested for other uses might avoid hair loss. Bald spots had an excessive amount of a protein called ?Prostaglandin D2? or PGD2, according to a study in Science Translational Medicine. Merck trial and error treatment for facial flushing and Actelion allergy combination, both in late- stage studies, block the healthy proteins.

About eight out of ten white men experience some balding before the age of 70, according to the researchers. Rogaine and Propecia slow the loss, and some users see minor hair growth. Men could probably re-grow all their hair if the inhibiting healthy proteins is removed, said George, chairman of dermatology at the University of Pennsylvania?s Perelman School of Medicine within Philadelphia.

We really do think if you take away the inhibition, you get longer hair, George, a study author, said in a telephone interview. We don?t know if the follicles will return to their former lengths, he explained. The study was funded by grants from the National Institutes of Health, the Pennsylvania Department involving Health, and other medical groups.

The researchers looked at the genes in the scalp samples from five men, comparing the bald parts to the haired sections. They found higher expressions of the gene that produces PGD2 in the bald samples, compared to the spots with hair. To be able as a guide, they found in samples of 17 men with hair loss that PGD2 was three times higher in the bald spots than where hair has been growing. The scientists then used mice to exhibit that excessive PGD2 decreased follicles.

Previous work has demonstrated that the stem cells that create hair continue to be intact in bald men, Cotsarelis said. The follicles are there, though they look smaller and produce finer, shorter hair. Over time, the hair is so short it no longer passes the top of skin.

New more-sensitive blood test catches recurring breast cancer a year earlier

ScienceDaily (Mar. 28, 2012) ? A new blood test is twice as sensitive and can detect breast cancer recurrence a full year earlier than current blood tests, according to a scientist who reported at the 243rd National Meeting & Exposition of the American Chemical Society (ACS) in San Diego on March 29.

Daniel Raftery, Ph.D., who reported on the test, pointed out that breast cancer survivors -- 2.5 million in the U.S. alone -- face about a 1-in-5 chance that the cancer will come back, or recur, within 10 years of treatment. Research shows that early detection of these recurrences and treatment can save lives. However, currently available blood tests are not very sensitive. Perhaps the best known test for a biological "marker" protein, or "biomarker," called CA 27.29, misses many cases of recurrence and detects them late -- often after symptoms, such as difficulty breathing or bone pain, surface.

"We have identified a group of nine biomarkers that signal recurrence of breast cancer," Raftery said. "Our markers detect twice as many recurrences as the CA marker does at the same specificity. They also detect cancer recurrence earlier, about 11-12 months sooner than existing tests. They accomplish this with blood samples, rather than biopsies, with less discomfort to patients."

To find these markers, Raftery's team at Purdue University and Matrix-Bio, Inc., a company he founded, analyzed many hundreds of "metabolites" in the blood of breast cancer survivors. Metabolites are small molecules, biological byproducts formed as the body's cells go about the business of life. Some are released into the bloodstream and urine. The rapidly emerging scientific field called "metabolite profiling" seeks to understand how these metabolites relate to health and disease. Groups of metabolites already have been linked to a range of diseases. Many of Raftery's biomarkers were known to be involved in cancer. But no one knew that this group of metabolites could serve as biomarkers for breast cancer recurrence, he said.

The markers are detected with an instrument called a mass spectrometer, which is common in clinical laboratories. Raftery explained that these markers would be used in combination with results from CA 27.29 blood tests.

"We take both of those results together and roll them into the profile so that the score we generate is a combination of the CA value and our nine metabolites," he said. "If the score indicates that the cancer probably has returned, the patient would then likely undergo imaging tests to locate the tumor."

Raftery hopes that the new test will become available later this year. In the meantime, the researchers are conducting another clinical study with the test. He also said that, in the future, the test might be useful in the early detection of breast cancer, not just recurrences.

The scientists acknowledged partial funding from the National Institutes of Health.

PROVIDENCE, R.I. [Brown University] If the fight against breast cancer were a criminal investigation, then the proteins survivin, HDAC6, CBP, and CRM1 would be among the shadier figures. In that vein, a study to be published in the March 30 Journal of Biological Chemistry is the police report that reveals a key moment for keeping cancer cells alive: survivin's jailbreak from the nucleus, aided and abetted by the other proteins. The research highlights that a protein's location in a cell affects its impact on disease, and offers clear new leads for the investigation.

All four proteins were already under suspicion. Researchers, for example, have already tried to assess what levels of HDAC6 in patients with estrogen-receptor positive breast cancer may mean for their prognosis. The results have been inconclusive. The new research suggests that measuring overall levels may not be enough, said the study's senior author Dr. Rachel Altura, associate professor of pediatrics in The Warren Alpert Medical School of Brown University and a pediatric oncologist at Hasbro Children's Hospital.

"We need to look not only at the levels, but also where is it in the cell," she said.

Altura's emphasis on location comes from what her research team found as they tracked and tweaked the comings and goings of survivin in cells. Inside the nucleus, survivin is no problem. Outside the nucleus, but within the cell, it can prevent normal cell death, allowing cancer cells to persist.

In previous work, Altura and her collaborators established that under normal circumstances, CBP chemically regulates survivin, a process called acetylation, and keeps it in the nucleus. The question in the new work was how survivin gets out.

In a series of experiments, what they observed was that in human and mouse breast cancer cells, HDAC6 gathers at the boundary between the nucleus and the rest of the cell, becomes activated by CBP, then binds survivin and undoes its acetylation. This deacetylation allows survivin to then be shuttled out of the nucleus by CRM1.

In the classic jailbreak, CBP is a corrupt guard who looks the other way as HDAC6, the shovel, is smuggled in. The final accomplice, CRM1, is the tunnel with a getaway car on the other end.

Working the new leads

Altura said the research suggests a clear strategy to keep survivin in the nucleus and two leads to pursue it, both of which she has already begun working on with collaborators in academia and in the pharmaceutical industry.

One idea is to inhibit HDAC6 in an attempt to prevent it from misregulating the acetylation of survivin. While general HDAC inhibitors are in clinical trials, Altura is optimistic that blocking just HDAC6, using specific inhibitors developed by a colleague in Japan, would have fewer complications.

"You always have to worry about all the things you don't know that you are targeting," she said. "If we can target HDAC6, we can maybe block survivin from coming out of the nucleus and maintain it in its good state."

The other strategy is to block CRM1, Altura said, an idea she is pursuing with a pharmaceutical company in breast cancer cells in the lab. She said preliminary experiments look promising in keeping survivin inside the nucleus and making cancer cells more susceptible to dying.

New evidence on effects of green coffee beans in weight loss

ScienceDaily (Mar. 27, 2012) ? Scientists have just reported striking new evidence that green, or unroasted, coffee beans can produce a substantial decrease in body weight in a relatively short period of time.

In a study presented at the 243rd National Meeting & Exposition of the American Chemical Society (ACS), the world's largest scientific society, Joe Vinson, Ph.D., and colleagues described how a group of overweight or obese people who consumed a fraction of an ounce of ground green coffee beans each day lost about 10 percent of their body weight.

"Based on our results, taking multiple capsules of green coffee extract a day -- while eating a low-fat, healthful diet and exercising regularly -- appears to be a safe, effective, inexpensive way to lose weight," Vinson said at the ACS meeting, being held in San Diego the week of March 26.? He is with the University of Scranton in Pennsylvania.

The study involved 16 overweight or obese people aged 22-26 years who took capsules of the extract or capsules containing a placebo, an inactive powder, for a total of 22 weeks. The subjects alternated between a low dose and a higher dose of the extract. The low dose consisted of 700 mg of the coffee extract, and the high dose was 1,050 mg. It was a so-called "cross-over" study in which people cycled through the two doses and the placebo, each for six weeks. Such studies have advantages because each person serves as his or her own "control," improving the chances of getting an accurate result.

All of the participants were monitored for their overall diet (calories, food eaten, etc.) and exercise over the study period. "Their calories, carbohydrates, fats and protein intake did not change during the study, nor did their exercise regimen change," Vinson said.

Participants lost an average of 17 pounds during the 22 weeks of the study. It included an average of a 10.5 percent decrease in overall body weight and a 16 percent decrease in body fat. Vinson noted that weight loss might have been significantly faster, except that participants received the placebo and the lower dose of green coffee extract for part of the study period.

Vinson pointed out that previous studies have shown weight loss with green coffee. But this was the first to use higher amounts of the coffee extract and the first to measure the response to various doses. Based on those studies, Vinson believes that green coffee beans' effects likely are due to a substance called chlorogenic acid that is present in unroasted coffee beans. Chlorogenic acid breaks down when coffee beans are roasted (usually at a temperature of 464-482 degrees Fahrenheit). Roasting gives coffee beans their distinctive color, aroma and flavor. Green coffee beans, in contrast, have little aroma and a slightly bitter taste.

When Apple announced the new iPad earlier this month, CEO Tim Cook claimed that the new iPad?s new A5X chip offers four times the performance of Nvidia?s quad-core Tegra 3 CPU. ?Now that the new iPad has hit the street, we?ve been able to compare its performance to that of an?Asus Eee Pad Transformer Prime, the first?tablet?with a Tegra 3 inside. The results may surprise you.

GLBenchmark 2.1
In GLBenchmark 2.1, ?one of the few benchmarks that has versions for both Android and iOS, the new iPad scored higher. On the program?s Egypt Standard test, which shows a 3-D animation of ancient Egypt, the new iPad and its A5X chip processed 6718 frames at a rate of 60 frames-per-second to the Transformer Prime?s 5,939 frames at 53 fps.?

When GLBenchmark ran the Geometric test (vertex weighted), which measures low-level shader performance, the new iPad processed?7,530,524 frames at a rate of 57 fps while the Tegra 3-based Transformer Prime processed just?3,523,926 at a rate of 27 fps. We saw the same trend on the program?s Fill test, which measures texturing speed, as the iPad processed?1.98 billion textels per second to the Prime?s 404.61 million.

Geekbench
On Geekbench, a cross-platform benchmark that measures raw processing power rather than graphics, the quad-core Tegra 3 blew its competitor way as it achieved an overall score of 1,571 to the A5X?s 692. On the integer (1391 to 614), floating point (2408 to 825) and memory subtests (1076 to 784) , the Tegra 3 dominated, but the A5X bested it by a small margin of 324 to 266 on the stream subtest.

Browser Benchmarks: Peacekeeper and Sunspider
We also tested the new iPad?s Safari browser performance on two synthetic benchmarks and compared the results to those on the Transformer Prime?s stock Android browser. In Peacekeeper, a general-purpose browser benchmark, the two tablets got nearly identical scores of 386 for the iPad and 387 for the Prime.?However, the iPad completed the Sunspider JavaScript rendering test in just 1810 milliseconds to the Prime?s 2216 ms.?

Unfortunately, since the two tablets use two different browsers, it?s impossible to isolate the processor?s role in giving the iPad slightly faster Sunspider results. It?s quite possible that Safari just performs better on this test than the Android 4.0 browser does.

Subjective Gaming Performance
Evaluating gaming performance on any tablet is a mainly subjective task, because there?s no benchmark that measures framerates like FRAPs does in Windows. However, we attempted to see what the differences in appearance were when we played the same game on both devices and placed them side by side.?

When we fired up the jet ski racing game "Riptide GP" on both devices, we noticed immediately that images looked sharper and more colorful on the new iPad?s retina display. Edges of objects like our avatar?s ski jacket, the walls on the racing course, and the writing on the scoreboard looked so much crisper on the iPad?s 2048 x 1536 screen and colors just popped a lot more than on the Transformer?s 1280 x 800 display. The difference in screen quality was only a little less pronounced when we played the third-person shooter game "Shadowgun."

However, when we looked past the beautiful screen, we noticed that the version of the game playing on the Transformer Prime had some additional visual effects that were not present on the new iPad. ?In "Riptide," we noticed more reflections in the water and a splashing effect that made it look like water was hitting the screen when our ski landed from a jump. On the iPad, we just saw a little foam appear on landing, not splashes.

In "Shadowgun," we walked into a scene with a burning helicopter and, on both devices, saw gorgeous billows of smoke and fire rising up to the sky. However, only on the Tegra 3-powered Transformer Prime did the game show two flags billowing in the wind; the flags weren?t even there on the iPad version.

Our experience playing "Shadowgun" and "Riptide" on the two tablets shows how difficult it is to separate processor performance from other system components like the screen. It?s likely that, if we could put a gorgeous 2048 x 1536 screen on the Transformer Prime, these games would have both the great special effects and the sharp, beautiful images.

However, comparing tablet-to-tablet rather than chip-to-chip, the iPad?s screen makes the gaming experience a much tastier visual feast than you get on the Transformer Prime. We hope that upcoming higher-res Android tablets like the 1920 x 1200 screened Transformer Prime TF701 will give mobile gamers the perfect combination of special effects and sharp, vibrant images.

It is also possible that, once developers rework their games for the new iPad?s A5X chip, we?ll see the same kind of added special effects that we see with Tegra 3. Neither "Shadowgun" nor "Riptide" has yet been optimized for the A5X chip, so who knows what they will look like in their next iOS iterations. For now, its clear that the A5X offers better benchmark performance, while Tegra 3-optimized games offer more and better visual effects.?

Breast cancer fundraising lags after abortion dispute

WASHINGTON (Reuters) - U.S. breast cancer charity Susan G. Komen for the Cure is feeling a pinch on donations following a controversy over its funding for Planned Parenthood, a leading provider of birth-control and abortion services.

A few of the group's flagship "Race for the Cure" fundraising events have failed to meet targets, a Komen spokeswoman said on Friday. Separately, at least five of the group's leaders have stepped down in recent weeks.

Komen, the world's biggest breast cancer charity, provoked uproar over its decision to cut - and later restore - funding for Planned Parenthood, a leading provider of birth control, abortion and other women's health services. Komen supports Planned Parenthood's efforts to provide access to breast-cancer screening.

The initial move to cut Planned Parenthood's funding became public in late January, and was viewed by some Komen supporters as a capitulation to political pressure from anti-abortion groups. Within a few days, the charity reversed course.

Komen said it had had problems meeting targets in about half of the five fundraising events it has staged since the blowup.

One in Lafayette, Louisiana, raised less than $400,000, below its $500,000 goal, and another, to be held in Fort Worth, Texas, is also struggling.

Komen spokeswoman Leslie Aun said the lower numbers appeared to stem from the controversy over Planned Parenthood.

"We're seeing challenges for races in some of our markets," Aun said. "People are concerned and they've decided not to help Komen. That's unfortunate because it affects women whose lives we're trying to help save."

FIVE MORE OFFICIALS STEP DOWN

Komen has said that in 2011 it screened 700,000 uninsured women for breast cancer, and it spends 83 cents of every donation dollar on research or community services.

Five Komen executives or directors have recently announced they are leaving the organization, although a group insider cited personal reasons for most of the resignations.

Nancy Macgregor, who has been with Komen since 1990, is leaving her role as vice president of global networks in June. Joanna Newcomb, director of affiliate strategy and planning, and Katrina McGhee, executive vice president and chief marketing officer, are also stepping down.

Aun said the chairman of Komen's board, LaSalle Leffall would resign his position to focus on a new role as provost at Howard University, though he will remain on the board. His replacement will be Robert Taylor, a Dallas attorney who had retired as Komen's founding board member in 2010.

Of the group's local leadership, Chris McDonald, head of the Komen affiliate in Oregon and Southwest Washington, announced on February 25 that she would resign. She said the controversy over Planned Parenthood affected her decision, but it was not the primary cause, according to a statement on the group's website.

Dara Richardson-Heron, the head of Komen's affiliate for greater New York City, announced on the group's website she was stepping down as "a personal decision."

The recent exodus follows the resignation of Karen Handel, a senior executive charged with spearheading the decision to cut funding for Planned Parenthood.

Some of Komen's members have also called for the resignation of founder Nancy Brinker, who created the organization in honor of her sister, who died from breast cancer.

Komen's board said it had "complete confidence" in the group's leadership.

"This isn't about Komen. This is about women," Aun said about the charity's work. "If people understand what's at stake, they'll come back and be supportive."

"Pariah," from first-time writer/director Dee Rees, doesn't break much artistic ground. It tells the same gay/lesbian coming-out story that we've seen a million times. But it's told particularly well and from within a black urban context, which I don't believe has been done before. It also goes a bit deeper into the hearts and minds of the homophobic parents than typically is done, which was great. Unfortunately, it only scratches those surfaces. Kim Wayans, who of course has a long history in comedy, shows she has major dramatic talent, playing the homophobic mother of the main character. The cast is universally good, but Wayans is the stand-out. The main character is a black teenage girl in Brooklyn going through the coming-out process. She has fully come out to herself as a lesbian, and she has even found her way into a lesbian circle of friends. She even frequents a women's night club. But she hasn't told Mom or Dad about any of this, both of whom are homophobic. Mom is particularly venomous in her hatred of gays and lesbians. You can see that Dad, a detective in the NYPD, in his heart of hearts is not a bigot. Thrown into the mix to complicate things a little bit is a bisexual girl eager to have lesbian experiences to explore herself. But she tosses lesbians aside like useless candy wrappers after she's had her fun. If I were going to give Dee Rees advice, I would say this: Ms. Rees, in "Pariah" you started digging into the parent characters with some real psychological and artistic depth. I encourage you to go more deeply in that direction. I think your true gifts as an artist lie there. I would give anything to see a sequel where you explore what happened to that mother and what she's really fighting. You hint that her husband is beginning to stray, but I think there's more in there. Help us see it. Remember when that great schoolteacher tells Alike that she could "go deeper" with her poetry? You could go deeper with your films. I know you could.
